LncRNA JADE is induced after DNA damage in an ataxia-telangiectasia mutated (ATM)-dependent manner. JADE is up-regulated in human breast tumors. Knockdown of lncRNA-JADE significantly inhibits breast tumor growth. Mechanistically, JADE transcriptionally activates Jade1, a key component in the HBO1 (human acetylase binding to ORC1) histone acetylation complex. Consequently, lncRNA JADE induces histone H4 acetylation in the DDR.

LncRNA JPH1-7 levels in head and neck squamous cell carcinoma (HNSCC) is up-regulated, and associated not only with poor prognosis, methyltransferase KMT2D (MLL1) mutation and concurrent TP53 mutation and 3p deletion, but also significantly correlated to advanced tumor stage. Furthermore, higher median JPH1-7 expression is observed among HPV-negative patients and patients reporting history of smoking. Knockdown of JPH1-7 reduces the expression of EMT-promoting genes for N-cadherin and Snail, as well as anti-apoptotic gene XIAP.
