LncRNA LUCAT1 can be induced by cigarette smoke extract (CSE) both in vitro and in vivo and is elevated in numerous lung cancer cell lines. It is located in chromosome 5 with four exons and three introns. The expression of LUCAT1 is regulated transcriptionally by nuclear factor erythroid 2-related factor (NRF2). A nuclear factor erythroid-derived 2 (NF-E2) motif is identified in the promoter region that shows binding to NRF2 after its activation. Functionally, the siRNA knockdown of LUCAT1 in human bronchial epithelial cells shows a significant potentiation of cytotoxicity induced by CSE in vitro (1).
Another study focusing on LUCAT1 reveals it is obviously up-regulated in non-small cell lung cancer tissues and its expression is associated with tumor size, tumor TNM stage, lymphatic metastasis and overall survival. LUCAT1 can significantly promote proliferation of lung cancer cells. Furthermore, LUCAT1 plays a crucial role in G1 arrest. What's more, LUCAT1 is correlated with the enhancer of zeste homolog 2 (EZH2) by repressing the expression of p21 and p57 (2).
