LncRNA MINCR has the highest and most significant positively correlated with MYC expression level. It has the MYC binding regions around its transcriptional start site (TSS) in burkitt lymphoma (BL) cells and P493-6 cells, with increasing peak intensity in MYC high conditions in P493-6 cells. Moreover, MINCR has all of the marks associated with open chromatin and active transcription in normal and malignant B cells. Besides, the expression of MINCR is reduced after MYC knockdown in BL cell lines. Functionally, MINCR knockdown is associated with impairment in cell cycle progression. Differential gene expression analysis after RNAi, shows among the genes down-regulated, a significant enrichment of cell cycle genes after MINCR knockdown. Interestingly, these genes are enriched in MYC binding sites in their promoters, indicating that MINCR acts as a modulator of the MYC transcriptional program. Accordingly, MINCR knockdown is associated with a reduction in MYC binding to the promoters of selected cell cycle genes. Finally, downregulation of Aurora kinases A and B and chromatin licensing and DNA replication factor 1 involve with the reduction in cellular proliferation observed on MINCR knockdown (1).
Besides, MINCR is also up-regulated in gallbladder cancer. It can promote gallbladder cancer cell proliferation and invasion (2).
