LncRNA NBAT1 is down-regulated in breast cancer and associated with tumor metastasis and poor patient prognosis. NBAT1 can inhibit migration and invasion of breast cancer cells. Mechanistic study shows that NBAT1 is associated with PRC2 member EZH2 and regulates global gene expression profile. Among them, DKK1 (dickkopf WNT signaling pathway inhibitor 1) is found to be regulated by NBAT1 in a PRC2-dependent manner, and responsible for NBAT1's effects on inhibiting migration and invasion of breast cancer cells (1).
While in neuroblastoma, CpG methylation and a high-risk neuroblastoma associated SNP on chromosome 6p22 functionally contribute to NBAT1 differential expression. Loss of NBAT1 increases cellular proliferation and invasion. It controls these processes via epigenetic silencing of target genes. loss of NBAT1 could affect neuronal differentiation through activation of the neuronal-specific transcription factor NRSF/REST (2).
Besides, NBAT1 is also down-regulated in renal cell carcinoma and correlated with histological grade, tumor stage, lymph node metastasis, and poor prognosis. NBAT1 promotes cell proliferation, migration and invasion (3).
